Alpha-1-antitrypsin
as a novel target for immunodeficiency
Alpha-1-antitrypsin as a novel target for human
immunodeficiency virus type 1 - target evaluation
and drug discovery strategies.
The endogenous serine protease inhibitor,
alpha-1-antitrypsin, represents a novel target for
the treatment of HIV-infection. Physiological
concentrations of alpha-1-antitrypsin inhibit HIV-1
production in chronically infected monocytes, block
cellular infection, and reduce virus replication in
freshly infected peripheral blood mononuclear cells.
Furthermore HIV-1 replication is considerably
enhanced in blood from individuals with hereditary
alpha-1-antitrypsin deficiency. A dossier has now
been described which:
> Analyzes the excellent proof of concept for
targeting serine protease as HIV therapies
> Establishes the pharmaceutical novelty of
serine proteases as targets for HIV infection
> Suggests screening architectures to develop
therapeutic candidates
> Identifies academic labs that could help
expedite screening or provide biological material
(such as human material) to fully characterize the
clinical potential of hits
