RWJ-270201, a once daily treatment of influenza About 10% to 20% of U.S. residents will get the flu each year. As a result an average of about 20,000 people per year will die and 114,000 per year have to be admitted to the hospital. Anyone can get the flu and serious problems from flu can happen at any age although the elderly, people with chronic medical conditions, and very young children are more likely to get complications from flu. Inoculation is currently the best prevention of flu however antiviral agents are in development. Amantadine and rimantadine are used to treat and prevent influenza A infections, zanamivir and oseltamivir, both neuraminidase inhibitors are used against influenza A and B infections. Zanamivir is inhaled, oseltamivir is taken orally and both require twice daily doses. Current challenges are to identify novel molecules with improved pharmacodynamics that would allow once daily oral dosing thus reducing cost. RWJ-270201 is a unique neuraminidase inhibitor with high selectivity for the influenza neuraminidase and with an efficacy comparable to or better than earlier neuraminidase inhibitors against a wide range of influenza A and B isolates.
