Oltipraz, a candidate antiangiogenesis molecule

Basic research efforts focusing on angiogenesis as a target for cancer therapeutics have yet to bear fruit. Of the 55 molecules in development, 43 are still at preclinical stages and considerable developmental activity is expected over the next decade. Oltipraz from Aventis is in clinical trials as an antischistosomal drug however it also appears to be an effective chemopreventative agent reducing the incidence of cancer. This is likely related to it's glutathione S transferase inducing activity. The anti-angiogenic properties of this molecule have now been investigated. Impressively Oltipraz reduced angiogenesis activity in vitro in human and rat assays. Furthermore in vivo, oral Oltipraz almost completely blocked the development of angiosarcoma xenografts. The observed efficacy of oltipraz in this model is comparable with that of SU 5416 and TNP-470, known antiangiogenic agents currently under clinical development. Oltipraz therefore represents an exciting anticancer candidate both through its well characterized chemopreventative activity and also through its more recently identified antiangiogenic activity.
