Angiogenesis inhibitors for the treatment of arthritis

Inhibitors of angiogenesis represent one of the highest profile classes of therapeutic candidates to have been developed over the past two decades. This therapeutic class is anticipated to represent a major focus of anti-cancer therapy and consequently the commercial rewards for developing a successful anti-angiogenic compound are immense. Indeed by 2006, the market for all products that play a role in angiogenesis is predicted to reach $1.75 billion. Although the spotlight has been on cancer in relation to this class, a rapidly growing body of evidence suggests that arthritis could be successfully treated with inhibitors of angiogenesis. Increased angiogenesis is a key component of synovitis and bone modeling in arthritis. Molecular mechanisms and pathological consequences of blood vessel growth in arthritis are now being elucidated. Preclinical studies of angiogenesis inhibitors in animal models of inflammatory arthritis support the hypothesis that inhibition of neovascularization may reduce inflammation and joint damage. Thus, companies involved in inflammatory disease, musculoskeletal disorders or vascular pharmacology should consider the increasing potential of angiogenesis inhibitors.
