BD BiosciencesCommonly referred to as a multiplexed bead assay, the Cytomtric Bead Array (CBA), is based on the use of a series of spectrally discrete particles that can be used to capture, (e.g. via an antibody) soluble analytes and then quantitate them. The analyte is measured by detection of a fluorescence-based emission and subsequent flow cytometric analysis.
The concentration of unknowns is calculated for the cytometric bead array in the same way as with any sandwich format assay, i.e. through the use of known standards and plotting unknowns against a standard curve.
The system thus generates data that are comparable to those from individual ELISA-based assays, but in a "multiplexed" or simultaneous fashion, since each analyte can be distinguished via the unique spectral characteristics of the particle.
Allowing quantitation of soluble analytes in samples never previously considered possible due to volume limitations, the system provides, in addition to the quantitative data, powerful visual images that can reveal unique profiles or protein signatures. The new BD Cytometric Bead Array (CBA) Flex Set system provides the reagents necessary for multiplex analysis, without any need for reagent formulation. The reagents are compatible with virtually all dual-laser flow cytometers capable of detecting and distinguishing fluorescence emissions at 576nm and 670nm. Automated sample acquisition and high throughput can be achieved with the 96-well plate based BD FACS array bioanalyser or plate loader equipped BD flow cytometers.
