InSite Vision

InSite Vision is an ophthalmic products company currently focused on ocular infection, glaucoma, and retinal disease. The Company specializes in three platform technologies: topical drug delivery (DuraSite), genomics, and retinal drug delivery. As a result, the Company has a portfolio of both diagnostic and therapeutic products in early and late-stage development. The Company's lead product, AzaSite™, targets infection of the eye, and will initially compete in the expanding market for ocular infection in the United States. AzaSite contains the drug azithromycin, a widely used, broad-spectrum antibiotic that has now been successfully formulated for topical application in the eye. AzaSite is formulated with DuraSite®, InSite Vision's patented drug-delivery vehicle, which offers the prolonged release of an active ingredient. This formulation could offer the benefit of a low-dosing regimen, attractive to both the eye-care patient and physician. The Company filed a New Drug Application in June 2006 and expects Inspire Pharmaceuticals, the AzaSite licensee for the U.S. and Canada, to commercially launch AzaSite in the United States in the second half of 2007. The Company also plans to pursue AzaSite license agreements outside of the United States during the second half of 2007. In the area of glaucoma, the Company conducts genomic research using the TIGR/MYOC gene, among other genes in the existing research/development portfolio. A portion of this research has been incorporated into the Company's OcuGene glaucoma genetic test for disease management. The commercially available OcuGene® test is a genetically based prognostic tool designed to detect a genetic marker (mt-1) in the promoter region of the glaucoma-related TIGR gene. Eye care specialists who utilize this test may gain valuable insight regarding disease management options, in combination with other risk factor data, before irreversible clinical changes become obvious, leading to diminished visual capability. Genomic research has also supported the development of ISV-205, the Company's novel glaucoma therapeutic. The ISV-205 formulation utilizes the DuraSite drug-delivery technology, and contains the drug diclofenac. The ISV-205 product candidate delivers concentrations of diclofenac that have been shown in cell culture systems to inhibit the production of the TIGR protein. Phase 2b clinical studies were successfully completed in 2001, and further development requires additional funding or collaboration with a strategic partner. Retinal development candidates include an intra-scleral delivery system device, represented as ISV-014. This device allows for the controlled and minimally invasive delivery of ophthalmic drugs to the retina and other tissues in the posterior chamber of the eye. The development of new opportunities related to the preservation and improvement of sight are directly associated with the Company's Mission and Vision Statement, designed to guide the activities and future direction of the organization.
