NEOKIMIA is a chemistry-based discovery company that integrates advanced synthesis and combinatorial chemistry capabilities with computational methods to produce libraries of novel, potentially bioactive molecules. The main component of the highly versatile, patented NEOKIMIA technology platform is the design and synthesis of conformationally defined, sophisticated macrocyclic molecules in a parallel fashion using (primarily) modern automated solid phase technologies. Appropriate recognition elements (small molecule building blocks, some of which are commercially available and others that are prepared internally) are assembled in a stepwise fashion together with a unique segment (a "tether" specially designed by NEOKIMIA) that restricts the molecule to specific spatial orientations. With the aid of computational techniques, the design of these macrocyclic libraries is based on consideration of diversity in terms of chemical composition, three-dimensional shape, and appropriate physical properties (Lipinski Rules of Five). To date, more than 35,000 macrocycles have been produced utilizing this proprietary approach. The versatility provided by this unique combination of diversity in terms of structure and conformation yields an extremely powerful discovery technique with applications in a wide variety of therapeutically relevant biological systems.
