    Poly(ADP-ribose) polymerases (PARPs) are a superfamily of 18 proteins, of which PARP-1 is the best characterized. PARPs are enzymes that are activated by DNA breaks to facilitate DNA repair

    Currently, there is a great deal of research being done on PARP, because its activity is linked to serious disease conditions, including cancer, ischemia, diabetes, arthritis, inflammatory bowel disease, and septic shock. Of greatest therapeutical interest are PARP-1 inhibitors, as they exert a duel effect of inhibiting cell death and reducing inflammation. Indeed, PARP inhibitors have been shown to be effective as radiation-sensitizing agents and appear promising in the treatment of conditions involving oxidative stress. Additionally, quantification of PARP activity and identification of abnormal patterns of PARP degradation by proteases might have utility in the diagnosis various conditions.
