Ezetimibe prevents the development of artherosclerosis In the USA, dietary cholesterol and saturated fatty acid intake is high significantly increases the risk of coronary artery disease. Since a staggering 12.4 million Americans suffer coronary artery disease identifying new hypolipidemic strategies is of vital importance. The high incidence of hyperlipidemia has created a market worth $15.9 billion world-wide and this has stimulated considerable pharmaceutical activity and consequently 182 molecules are in various stages of development or on the market. 43% of these are in preclinical development suggesting that interest in this field is stable and hence the search for new candidates continues. Schering-Plough's Ezetimibe, represents the first selective inhibitor of intestinal cholesterol absorption. Cholesterol absorption has been shown to be inhibited by >90% at doses of ezetimibe >3 mg/kg in apoE-/- mice which represent a model of atherosclerosis. These animals fed a western diet develop aortic atherosclerotic lesions which are reduced by 80% following drug treatment. Similarly, carotid artery atherosclerotic lesion cross-sectional area was reduced by 97%. This data confirm that Ezetimibe is able to reduce cholesterol levels and for the first time shows, crucially that it can reduce the development of atherosclerosis.
