Reversing tumor drug resistance A variety of chemotherapies are available to oncologists and these generally reduce the rate of tumor progression. However intrinsic or acquired tumor-mediated drug resistance is a major clinical obstacle that can result in the lack of tumor responsiveness in patients undergoing treatment. Drug resistance is in part due to active efflux transporters such as p-glycoprotein and consequently the development of inhibitors of this protein represents an active area of the pharmaceutical industry. With this challenge in mind, researchers at The Kitasato Institute in Tokyo have synthesized a series of new isoprenoid derivatives. The lead compound, N-5228 bound to p-glycoprotein and potentiated the accumulation of vincristine in drug-resistant cells. This candidate was therapeutically active in its own right demonstrating efficacy in an in vitro assay of human MDR-type resistant carcinomas. N-5228 was also tested in a xenograft model and demonstrated anti-tumor activity in mice bearing MDR-type resistant tumors. This series thus has anti-cancer potential and further development is eagerly awaited.
