5-HT7: Pharmaceutical opportunities in the fields of CNS and cardiovascular disorders

E Sussex, UK 15th January 2003 --- New LeadDiscovery Dossier. 5-HT7 receptors are implicated in the pathogenesis and prophylactic treatment of migraine as well as depression, schizophrenia and cardiovascular disease. This DiscoveryDossier provides a general description of the 5-HT receptor family, with an emphasis on the current understanding of the 5-HT7 receptor, overviewing its expression profile and physiological roles. The report continues with a description of the role that 5-HT7 receptors play in the pathophysiology of depression, schizophrenia and cardiovascular disease. The report then focuses on migraine, a market that has expanded five-fold since the 1990s due to an increase in awareness and therapeutic options. The development of 5-HT1D agonists has contributed to both of these aspects of migraine therapeutics. This dossier gives a full analysis of the migraine arena describing emerging and established treatment options and then discusses the proof of concept supporting the development of 5-HT7 receptor antagonists as improved antimigraine treatments. The report concludes with an assessment of patent activity relating to 5-HT7 and a discussion of the tools available to researchers wishing to develop 5-HT7 receptor antagonists. Depending on the level of interest an overview of medicinal chemistry surrounding the development of these molecules, proposed strategies and also in silico derived libraries of candidate 5HT-7 receptor antagonists can be provided. For companies wishing to enter into or expand 5HT-7 screening activities LeadDiscovery in collaboration with Chemovation Ltd can synthesize and provide samples of molecules from these libraries.
