Summary of "Natural Products for Drug Discovery: Discovery of Gramines as Novel Agents against a Plant Virus."

Plant viral diseases cause tremendous decreases in yield and quality. Natural product gramine (1) and its structure simple analogues 2-35 were synthesized from indoles, amines and aldehydes in one step. The antiviral activities of these alkaloids were evaluated for the first time. Most of these compounds displayed higher antiviral activities than commercial antiviral agent ribavirin. Compounds 22, 30 and 31 exhibited significantly higher antiviral activities than ningnanmycin (perhaps the most successful registered antiplant viral agent), emerging as new lead compounds for antiviral research. In vitro capsid protein (CP) assembly and Tobacco Mosaic Virus (TMV) rod assembly inhibition determinations indicated that these compounds likely exerted their virus inhibition by crosslinking TMV CP and interfered with virus assembly. Further fungicidal activity tests against 14 kinds of phytopathogenic fungi revealed that gramine analogues displayed broad-spectrum fungicidal activities. Although gramine has been reported to have influence on germination and development of Erysiphe graminis, all of the compounds showed no inhibitory effect against Blum eria graminis f. sp.tritici on wheat in our test. Some of these compounds also exhibited certain insecticidal activities.
