Summary of "Hitherto-unexplored Photodynamic Therapy of Ag2S and Enhanced Regulation Based on Polydopamine in vitro and vivo."

Given the superior penetration depth, photosensitizer with longer absorption wavelength presents broader application prospect in photodynamic therapy (PDT). Herein, Ag2S quantum dot was discovered, for the first time, to be capable of killing tumor cell in the photodynamic way by near-infrared light irradiation, which meant relative less excitation of probe compared with traditional photosensitizer absorbing short wavelength. With further modification of polydopamine (PDA), the outstanding capacity of PDA-Ag2S inducing reactive oxygen species was obtained which increased by 1.69 times. With the regulation of PDA, more opportunities were provided for Ag2S to react with surrounding O2 demonstrated by the typical triplet electron spin resonance (ESR). Furthermore, the PDT effects of Ag2S and PDA-Ag2S achieved at longer wavelength were almost identical to the effects produced at 660 nm proved by studies in vitro. And distinctly better therapeutic phenomenon of PDA-Ag2S than Ag2S was shown in experiments in vivo, which further validated the enhanced regulatory effect of PDA. Together, a new photosensitizer with longer absorption wavelength was developed, with the hitherto-unexplored photodynamic function of Ag2S quantum dot extended and its enhanced regulatory effect originated from PDA.
